چکیده

Supplemental Figures S2-S6 show how three characteristics of the dorsal midline peak of [LR] vary when several of the critical parameters are altered, pairwise, over a substantial range from the “base” parameter set of Figure 5. In each figure, the top left image depicts the peak height at 38 min, the time given in Figure 6. The top right image depicts peak height at 90 min. This was used, rather than steady-state peak height, because for some parameter sets, steady state occurs later than 90 min, a time too long to be biologically significant. The lower left image depicts the time required for the height of the [LR] peak to achieve 63.2% (1-e) of its 90 min value; this gives a sense of the overall rate of formation of the BMP activity gradient. The point at the very center of each square represents the parameter set used is Figure 5.

Synthetic enzyme-substrate tethering obviates the Tolloid-ECM interaction during Drosophila BMP gradient formation

Members of the Tolloid family of metalloproteinases liberate BMPs from inhibitory complexes to regulate BMP gradient formation during embryonic dorsal-ventral axis patterning. Here, we determine mechanistically how Tolloid activity is regulated by its non-catalytic CUB domains in the Drosophila embryo. We show that Tolloid, via its N-terminal CUB domains, interacts with Collagen IV, which enhan...

Stepwise formation of a SMAD activity gradient during dorsal-ventral patterning of the Drosophila embryo.

Genetic evidence suggests that the Drosophila ectoderm is patterned by a spatial gradient of bone morphogenetic protein (BMP). Here we compare patterns of two related cellular responses, both signal-dependent phosphorylation of the BMP-regulated R-SMAD, MAD, and signal-dependent changes in levels and sub-cellular distribution of the co-SMAD Medea. Our data demonstrate that nuclear accumulation ...

Dorsoventral Polarity of the Nasonia Embryo Primarily Relies on a BMP Gradient Formed without Input from Toll

In Drosophila, Toll signaling leads to a gradient of nuclear uptake of Dorsal with a peak at the ventral egg pole and is the source for dorsoventral (DV) patterning and polarity of the embryo. In contrast, Toll signaling plays no role in embryonic patterning in most animals, while BMP signaling plays the major role. In order to understand the origin of the novelty of the Drosophila system, we h...

Formation of the BMP activity gradient in the Drosophila embryo.

The dorsoventral axis of the Drosophila embryo is patterned by a gradient of bone morphogenetic protein (BMP) ligands. In a process requiring at least three additional extracellular proteins, a broad domain of weak signaling forms and then abruptly sharpens into a narrow dorsal midline peak. Using experimental and computational approaches, we investigate how the interactions of a multiprotein n...

BMP Signalling: Synergy and Feedback Create a Step Gradient

More than a decade ago, genetic evidence predicted the existence of a Dpp gradient in the early Drosophila embryo. Two recent studies finally reveal Dpp distribution, providing further insights into the mechanism of BMP gradient formation.
